生殖器疱疹患者的单纯疱疹病毒PCR实验诊断研究

摘要 目的 探讨对生殖器疱疹患者进行单纯疱疹病毒PCR实验诊断的必要性和实用性。方法 采用聚合酶链反应(PCR)对生殖器疱疹患者组 82例及对照组 30例进行 HSV检测。结果GH患者组检出阳性75例,占91.46%,检出率明显高于对照组(P<0.01)。结论 PCR作为HSV感染的实验诊断技术具有很好的应用价值。 Objective To study the detection of herpes simplex virus DNA for the diagnosis of genital herpes by polymerase chain reaction. Methods The polymerase chain reaction was used for rapid and specific detection and typing of different viruses. We selected one primer pair from the HSV DNA polymerase coding gene to detect and type HSV from virus isolates.The primers bracket a 229 base pair fragment of the HSV- 1 genome and a 241 base pair fragment of the HSV 2 genome.The polymerase reactions were carried out in 0.5ml microcentrifuge tubes with a final volume of 25ul.The amplification consisted of 32 cycles at 95℃ for 1min(denaturation), 55# for 1min(annealing)and 72# for 1min(extension). After PCR the type of HSV present was determined on the basis of size of the ethidium bromide stained band by agarose gel elec-trophoresis.The specimens were derived from clinical samples of diagoscd as GH(82 cases)and control group(30 cases) . Results Among 82 cases of GH patients, the general positive rate of HSV was 91.46%, the positive rate of the control group was 6.67% .The rate of positive detection of HSV in the GH patients was higher than that in the control group. Conclusion The simple and rapid PCR assay is specific and sensitive for HSV detection. HSV genotyping by PCR has diagnostic significance for the clinical practice and it is of importance for cpidemiological reasons.
