摘要
目的 研究穿孔素对不同肿瘤细胞株的杀伤作用是否一致 ,以确定肿瘤细胞对穿孔素的杀伤是否有抵抗作用。方法 大量培养LAK细胞 ,制备含穿孔素毒性颗粒的LAK细胞裂解物 ,用红细胞裂解试验证实LAK细胞裂解物的杀伤活性。培养穿孔素敏感的K5 6 2肿瘤细胞株和穿孔素不敏感的Hela、MGC80 3肿瘤细胞株 ,用流式细胞计数的方法研究LAK细胞裂解物对3种不同肿瘤细胞的杀伤作用。结果 红细胞裂解试验证实 ,LAK细胞裂解物具有杀伤活性 ;抗人穿孔素 (Humanperforin ,HP)McAbs可封闭LAK细胞裂解物对不同肿瘤细胞株的细胞毒作用 ;LAK细胞裂解物对K5 6 2细胞的杀伤是通过穿孔素依赖途径的。结论 穿孔素对K5 6 2、Hela和MGC80 3肿瘤细胞的杀伤明显不同 ,说明肿瘤细胞存在对穿孔素的抵抗机制。
Objective To study whether the killing effects of human perforin on various tumor cells and to determine whether tumor cells can resist human perforin. Methods The hemolytic assay was used to detect the cytolytic activity of the lysate of LAK cells and the prepared McAbs were used to block the cytolytic activity.The flow cytometer was used to analyze the cytotoxic activity of perforin to different tumor cells lines.Results Hemolytic assay showed that the prepared McAbs could block the cytolytic activity of LAK lysates.The flow cytometer analysis showed that the perforin sensitive K562 cells could be killed by LAK lysates and the cytolytic activity of LAK lysates could be blocked by the prepared McAbs;the Hela cells and MGC80-3 cells could not be killed by LAK lysates.Conclusion Tumor cells are resistant to human perforin.
