神经保护蛋白同源基因在阿尔茨海默病大鼠海马中的表达 被引量:1

摘要 目的 探讨活性依赖性神经保护蛋白 (activity -dependentneuroprotectiveprotein ,ADNP)同源基因的表达与阿尔茨海默病的关系。方法 采用淀粉样β 肽注射大鼠脑室 ,制备阿尔茨海默病样损伤大鼠模型 ,15d后通过RT PCR方法检测大鼠脑海马胆碱乙酰转移酶 (cholineacetyltransferase,ChAT)基因、活性依赖性神经保护蛋白基因及Mn -SOD、Cu ,Zn -SOD基因的表达。结果 胆碱乙酰转移酶、ADNP基因的表达在实验组海马中均下降 ,Cu ,Zn -SOD基因的表达没有明显变化 ,而Mn -SOD则有所升高。结论 结果提示ADNP表达降低 ,使神经细胞缺乏保护因素 ,与阿尔茨海默病的发生发展相关。 Objective To investigate the interrelation of the expression of activity-dependent neuroprotective protein (ADNP) homogenetic gene and Alzheimer's disease.(AD).Methods The rat models of Alzheimer's disease were prepared with infusion of amyloid beta-peptide into the cerebral ventricle in 8?week-old rats.15 days later,the expressions of choline acetyltransferase (ChAT),activity-dependent neuroprotective protein, Cu,Zn-SOD were determined with the methods of RT?PCR.Results The expressions of choline acetyltransferase and activity-dependent neuroprotective protein were decreased in the hippocampus of experimental rat group, while the expression of Mn-SOD was increased, but no change of Cu,Zn-SOD.Conclusion The results suggest that the decreasing of ADNP might be interrelated with Alzheimer's disease.
