摘要
目的 研究脑室注射Aβ1 40 对海马NAPOR基因表达的影响。方法 利用立体定向仪将Aβ1 40 注射脑室 ,建立大鼠AD模型 ,采用逆转录PCR方法研究NAPOR在海马的差异表达。结果 通过莫氏水迷宫实验及胆碱乙酰转移酶的基因表达验证AD模型成功 ,NAPOR的RT PCR结果表明该基因片段在AD鼠海马高表达。结论 NAPOR为RNA结合蛋白 ,其表达的时空模式与神经元凋亡密切相关。结果提示Aβ1 40 对海马神经元的毒性可能通过改变NAPOR基因的表达 ,促进神经元凋亡而实现。
Objective To study the effects on the gene expression of NAPOR in hippocampus of rat by injection of Aβ 1?40 to brain ventricle.Methods The model of Alzheimer's disease of rat was established by injection of Aβ 1?40 to brain ventricle.RT?PCR technique was used to detect the differential expression of NAPOR in hippocampus.Results The model was verified successfully by Morri's water maze and expression of gene ChAT.The data of RT?PCR for NAORP revealed that it had higher expression in hippocampus of AD rat.Conclusion NAPOR is a RNA?binding protein,its expression pattern is spatially and temporally coincident with neuronal apoptosis.The results suggest that toxicity of Aβ 1?40 to neurons may result from alteration of NAPOR expression and further promote apoptosis of hippocampus neuron.
