祛风合剂对Ⅰ型变态反应影响的实验研究

Experimental Study of Influence of "Wind-Dispelling Mixture" on Type I Allergic Reaction

为研究祛风合剂抗Ⅰ型变态反应的药理作用 ,进行小鼠耳异种被动皮肤过敏试验(PCA)、大鼠颅骨骨膜肥大细胞脱颗粒试验、大鼠组胺诱发的足跖肿造型试验 ,观察祛风合剂大、小剂量和西药 (异丙肾上腺素、异丙嗪 )对其影响。结果 :大、小剂量祛风合剂均对小鼠耳异种被动性皮肤过敏有明显的抑制作用 ,且其作用与异丙肾上腺素相似 ;大、小剂量祛风合剂、异丙肾上腺素都能抑制肥大细胞脱颗粒 ,但祛风合剂作用弱于异丙肾上腺素 ,且大、小剂量组之间无明显差异 ,P>0 0 5;光镜下肥大细胞形态学呈现类似变化 ;祛风合剂能抑制由组胺诱发的足跖肿 ,但作用比异丙嗪弱 ,在 3 0min和 2 4 0min时 ,小剂量组优于大剂量组 ,P <0 0 5。提示祛风合剂可抑制抗原与特异性IgE结合 ,抑制肥大细胞脱颗粒及抗组胺 ,从而起到抗Ⅰ型变态反应作用。

To investigate the pharmaceutical effect of 'Wind-Dispelling Mixture'on type I allergic reaction,passive cutaneous anaphylaxis test in mice,mastocyte degranulation test of pericranium in rats,histamine-induced metatarsal swelling test in rats were performed to observe the effects of 'Wind-Dispelling Mixture',isoproterenol and phenergan on them.Results showed that large and small dosage of'Wind-Dispelling Mixture'could obviously inhibit the cutaneous anaphylaxis,similar to isoproterenol;large and small dosage of'Wind-Dispelling Mixture'and isoproterenol could inhibit the degranulation of mastocyte,and with a weaker function of the'Mixture'than isoproterenol,but there was no difference between the large dosage and small dosage( P >0 05);light microscope showed similar changes;the 'Mixture'could ihhibit the metatarsal swelling,but weaker than phenergan;at the moment of 30 minutes and 240 minutes after experiment,the effects showed that small dosage was better than large dosage( P <0 05).It is revealed that'Wind-Dispelling Mixture'can inhibit the combination of antigen and IgE,degranulation and mastocyte and histamine,eventually to suppress type I allergic reaction.
