摘要 探讨细胞周期调控蛋白CyclinD1,CyclinE ,Cdk2 ,Cdk5在K5 6 2细胞增殖和分化时的不同表达。方法 :本实验采用Western -Blotting -ECL方法分析了苦参碱作用K5 6 2细胞前、后 2 4,48,72h ,CyclinD1,Cy cliE ,Cdk2 ,Cdk5的不同表达。结果 :在增殖的K5 6 2细胞中 ,伴随着DNA合成增加 ,CyclinE和Cdk2保持高表达 ;而在分化的细胞中 ,随着DNA合成减少 ,CyclinD1,Cdk5表达增强。结论 :在细胞从增殖向分化途径逆转时 ,伴随着一些特殊的Cyclin/Cdk表达的改变。 Objective:To investigate the differential expression of proteins regulating cell cycle progression such as CyclinD1,CycinE,cdK2 and cdK5 in K562 cell line.Methods:By Western-Blotting-ECL analysis,CyclinD1,CycinE,cdK2 and cdK5 were detected in k562 cells untreated or treated and with matrine for 16h,24h,48h.Results:The results showed that CyclinE and cdK2 were expressed at relatively high levels the nuchlei of prolifeation cells,accompanied by increased DNA synthesis,whereas CyclinD1 and cdK5 were expressed at comparably high levels in the nuclei of differentiation-induced cells,correlated with decreased DNA synthesis.Conclusions:It suggests that in K562 cells,the progression from growth to differentiation,is accompanied by the differential expression of sepcific cyclin/cdk pairs,comprising cdk2/cyclinE in growth and cdk5/cyclinD1 in differentiation.
