摘要
目的:从凋亡之异常角度探讨儿童髓母细胞发生、发展及预后的生物学机制。方法:以84例获随访的儿童髓母细胞瘤组织(按术后生存期3、5、10年分为A、B、C三组)为研究对象,用原位杂交及免疫组化染色法分别检测Bcl-2mRNA和增殖细胞核抗原(PCNA)的表达,并用31末端标记法做原位细胞凋亡检测。结果:72例(85.7%)表达Bcl-2mRNA,其阳性率为A>B>C;PCNA阳性肿瘤组织细胞密度与肿瘤细胞Bcl-2mRNA表达水平增加,呈正相关,而凋亡肿瘤细胞密度与肿瘤细胞Bcl-2mRNA表达水平呈负相关。结论:儿童髓母细胞瘤中Bcl-2基因高表达可抑制其凋亡,细胞增殖与凋亡失衡可能是儿童髓母细胞瘤发生发展的重要环节。
To study the biological mechanism of progression and prognosis of medulloblastoma in children, based on abnormal apoptosis. Method: The expression of Bel - 2 mRNA, proliferating cell antigen (PCNA) and the apoptosis of medulloblastoma from 84 children with 3,5,10 survival years (group A, B, C), respectively were studied with in situ hybridization and immunohistochemistry, and in situ death detection (Tunel method) . Result: Positive expression rate of Bel - 2mRNA was 85.7%(72 cases) , and its order was group A > B > C among three group. With the increased expression of Bel - 2mRNA, the cell density of positive PCNA increased, but apoptosis decreased in the medulloblastoma. Conclusion: Over ?expression of Bel - 2 mRNA inhibited apoptosis of medulloblastoma in children. The imbalance between cell proliferation and apoptosis maybe a key to formation and progression of the medulloblastoma in children.
