摘要
目的 :通过不同浓度的他莫昔芬 (TAM)对体外培养的MCF - 7细胞周期进程的影响 ,探讨TAM治疗乳腺癌的作用机制。以便为MCF - 7细胞同步化后 ,再应用化疗药物 ,为临床治疗乳腺癌奠定理论基础。方法 :采用人乳腺癌细胞MCF - 7体外培养 ,加入不同浓度的TAM(即 0 1nm ,10nm ,10 0nm和 1μm)培养一定时间后 ,检测其细胞周期各时相的变化。结果 :发现随TAM浓度的增高 ,MCF - 7细胞G0 /G1期不断增高。结论 :TAM可抑制MCF - 7细胞周期进程。TAM浓度在 1μm时 ,89 6 %的MCF - 7细胞集中在G0 /G1期。
Objective The effect of different dose of tamoxifen(TAM) on the growth of human breast cancer cell line MCF-7 in cell culture in vitro has been observed,indicated that TAM may inhibit the growth process of MCF-7 cell.It is showed that utilization of cell cycle synchronization in clinic may enhance the effect of chemotherapy on breast cancer.Method During cell culture in vitro of breast cancer cell MCF-7,adding different dose of TAM in media (0 1nm,10nm,100nm and 1μm) calculating cell ratio in each stage.Results The ratio of cancer cell in G 0/G 1 stage increased along with increasing dose of TAM in media.Conclusion TAM may inhibit the growth process of human breast cancer cell line MCF-7.When concentration of TAM is lum in media,almost 89 6% of cancer cell stay in G 0/G 1.
