SmIgM^+与SmIgG^+B细胞对EB病毒转化敏感性的比较

COMPARISON OF SUSCEPTIBILITES OF SMIGM^+ AND SMIGG^+ B LYMPHOCYTES TO EBV TRANSFORMATION

摘要 EB病毒(EBV)转化的B细胞具有优势分泌人IgH的特点。本文应用Panning法分离正常人外周血中SmIgM^+和SmIgG^+B细胞亚群,分别用EBV转化,再与人-鼠骨髓瘤细胞系SHM-D33融合,以转化率和融合率两个指标比较两种B细胞亚群在转化和融合水平的差异。结果表明,SmIgM^+B细胞亚群对EBV转化的敏感性是SmIgG^+B细胞亚群的4~5倍:而融合率无显著差异。部分解释了EBV转化和EBV-杂交瘤技术中人IgM优势分泌现象的原因。所采用的Panning技术还可用于定向生产IgG类人单抗。 EB virus(EBV) transformed human B lymphocytes dominantly secret human IgM. In this article, SmIgM+ and SmIgG+ B lymphocyte subsets isolated from human peripheral blood with 'Panning' technique were transformed by EBV, then fused with a human-mouse myeloma cell line SHM-D33 respectively. The differences in EBV transformation and hybridization were observed. The results indicated that the susceptibility of SmIgM+ B subset to EBV transformation was 4-5 times as many as that of SmIgG+ subset, whereas the fusion rates of these two subsets had no marked differences. Thus, the phenomena of dominant human IgM secretion in EBV transformation and EBV-hybridoma technique could be partly explained.The 'Panning' technique adapted can also be applied for producing human monoclonal antibodies of IgG class.
