摘要
目的 研究对比人血白蛋白及四氯化碳 ( CCl4 )两种制备大鼠肝纤维化模型的差别 ,为临床实验研究提供参考。方法 健康 SD大鼠分别用 CCl4 皮下注射法和人血白蛋白免疫法诱导肝纤维化模型 ,观察大鼠生存率、体重、肝功能、血清 PC 、C 、HA水平和肝组织病理学变化情况。结果 白蛋白造模大鼠平均体重较 CCl4 模型大鼠高 ,但动物死亡率高 ,肝纤维化形成率低 ,平均纤维化积分较低 ,与 CCl4 模型比较差异显著 ( P<0 .0 5 )。结论 CCl4 皮下注射法造模简便易行、价格低廉、动物死亡率低、模型成功率高、病变典型 ,可作为肝纤维化实验研究的首选模型。
Objective To investigate and determine the difference of human albumin induced and CCl 4 induced liver fibrosis in rats.Methods Male SD rats were divided into 3 groups:normal(N),human albumin model(A),and CCl 4 model(C).The rats of group C were given hypodermic injection of 30% CCl 4(3ml/kg,2times/week)for 16 weeks;and the rats of group A were given intravenous injection of human albumin.The liver samples were subjected to histopathological examination,and the changes of hepatic content of hydroxyproline and serum PCⅢ,CⅣ,HA,AST,ALT,TP,A,TB were observed.Results The average death rate and the weight of rats were higher in group A than in group C.However,the rate of liver fibrosis and serum levels of PCⅢ,CⅣ,HA were lower in group A.Conclusion CCl 4 induced liver fibrosis in rats is a cheaper,easier,reliable method and has practical usefulness in clinical researches.
