负压对肢体缺血犬血浆内皮素的影响

摘要 目的 观察肢体负压 (LNP)对肢体动脉闭塞犬血浆内皮素 (ET)的影响 ,并探讨其对ET影响的意义。方法 犬 2 0只 ,随机分治疗组 10只和对照组 10只 ,均采用切断左后肢股动脉所有分支 ,动脉腔内置入一根螺旋状金属丝的方法 ,制作肢体动脉闭塞性病变模型。在模型制作后 2周 ,治疗组行患肢负压治疗 ,连续 10d ;对照组不做负压治疗。两组均于模型制作前、制作后 2周及负压治疗 10d后或相应时间 ,用放射免疫分析的方法检测患肢股静脉血浆ET水平 ,治疗组随机选 4只犬 ,分别于负压治疗第 1,5 ,10 ,15天 ,测血浆ET水平。结果 负压治疗后 ,治疗组血浆ET水平显著低于治疗前 ,由治疗前 (112 .2 5± 10 .14) pg/ml降低到治疗后 (81.0 2± 7.84) pg/ml,P <0 .0 0 1。随着治疗时间延长 ,浓度逐渐减低 ,对照组LNP相应时间ET无明显变化 (P >0 .0 5 )。结论 肢体负压可显著降低肢体动脉闭塞犬的血浆ET水平 ,且随治疗时间延长逐渐降低。 Objective To observe the effect of limb negative pressure (LNP) on plasma endothelin (ET) in dogs with arterial occlusive disease(AOD) and explore the significance of effects of LNP on ET.Methods Twenty dogs were divided randomly into the treatment group(10 dogs) and the control group(10 dogs). By cutting the all branchs of femoral artery and inserting a piece of spiral wire into the artery of canine left posterior limb, we established the animal model with extensive arterial occlusive disease of extremity. The operated limbs of the treatment group were given LNP therapy while those of the control group were not. At the respective stages of pre-model, 2 weeks after the modeling and LNP treatment completion,we determined plasma ET with radioimmunoassay(RIA). Also,we detected plasma ET of those four dogs selected randomly, in 1,5,10,15day after the treatment respectively.Results After LNP treatment, plasma ET decreased significantly from (112.25±10.14)pg/ml to (81.02±7.84)pg/ml,P<0.001.No significant difference (P> 0.05) was found in index in the control group. Conclusion LNP may significantly decrease plasma ET of ischemic extremity and drop it gradually according to the treatment.
