脱氧雪腐镰刀菌烯醇(Deoxynivalenol)与2,4-二氯苯氧乙酸之间在小麦愈伤组织分化、植株K^+吸收和运输上的拮抗

摘要 将小麦品种徐州21的愈伤组织置于MS培养基于光下培养,以加2ppm 2,4-D于MS基本培养基为对照,设三个浓度(10^(-6),10^(-5),10^(-4)M)的三种禾谷镰刀菌(Fusarium graminearum schw.)毒素Deoxynivalenol(简称DON)、T_2和玉米赤霉烯酮九个处理。另以10^(-6),10^(-5),10^(-4),10^(-3),5×10^(-3)M五个DON粗毒素浓度(经高效液相标定)作对比。30天后,经DON处理的愈伤组织都出现根分化,地上部分分化不明显,且粗、纯品效应相同。在10^(-6)—10^(-3)M DON浓度下,根分化量随浓度提高而增多。5×10^(-3)M DON粗毒素处理的愈伤组织大部分变褐坏死,但仍有较多根分化。而T_2和玉米赤霉烯酮与对照一样,无明显分化。说明DON能拮抗2,4-D的抑制分化效应。为搞清拮抗性质,我们对在0.2mM CaSO_4中水培5天的扬麦5号幼苗进行根系处理,36小时后进行K^+吸收和运输分析。结果表明10^(-5)M_2,4-D、10^(-3)M DON及10^(-5)M_2,4-D+10^(-3)M MS tissus culture medium containing only 2ppm 2,4-D was as control, different concentrations of three Fusarium toxin Deoxynivalenol (DON), T2 and Zearalenone was put into medium. After 30day's culture under light, roots were differentiated from those callus treated by 10^(-6)M, 10^(-5)M, 10^(-4)M, 10^(-3)M, 5×10^(-3)M DON. But no differentiation was observed in treatment of T2, Zearalenone and the control. It indicated DON antagonized 2,4-D effect by inhibiting differentiation. Minewhile, the antagonism between DON and 2,4-D also occurred in root K^+absorption and translocation from shoot to root in wheat.
