摘要
以日本京都大学采自拉美各国的部分野生辣椒为材料,共计5个种、9个变种:C.annuum,var.annuum,C.annuum,var.mininum,C.baccatum,var.baccatum,C.baccatum,var.pendulum,C.chinense,C.frutescens,和 C.pubescens。愈伤组织诱导培养基以 MS 附加2.0mg/l NAA,0.3mg/l 6BA 较好。其中,C.baccatum,var.baccatum 和可栽培利用的 C.annuum,var.annuum 的子叶愈伤组织在 MS+0.1mg/l NAA+0.05mg/l IAA+6.6mg/l 6BA,以及 MS+0.4mg/l NAA+0.1mg/l IAA,0.1mg/l 6BA 分化培养基上均能获得再生绿苗。
Cotyledon hypocotyl and root excised from sterile seedlings germinated in hormone-free MS medium in nine strains of both cultivated species and wild species in Capsicum,i.e.C.annuum,var.annuum,var.mininum,C.baccatum,var.baccatum. C.baccatum,var.pendulum,C.chinence,C.frutescens and C.pubescens,were imp- lanted onto MS medium with different hormone combinations and different concentra- tions on each several hormone alone.The callusing response of such explants was tested on 17 kinds of callus induction medium,in which D4 medium,MS medium supplemented with 2.0mg/l NAA and 0.3mg/l 6BA was the best of all for inducing to- tipotent callus in quantity,organogenesis was observed and a total of 13 plantlets and shoots arising from C.annuum,var.annuum and C.bacatum,var.baccatum were rege- nerated on media with NAA and 6BA in different combinations,such as 0.1mg/l NAA plus0.6mg/l 6BA,or 0.4mn/l NAA plus 0.1mg/l 6BA and such like.The effect of IAA which had been emphasized in another culture of pepper plant was not so impor- tant in our experiments. Up to three shoots could appear in a single differentiating callus. Cytological observation in regenerating root tip indicated a diploid chromosome number(2n 24)in the majority of root tip cells.
