大骨节病病儿血浆致培养兔软骨细胞损伤实验的形态学观察

A morphological observation on damage experiment to cultural rabbit chondrocytes caused by blood plasmas of KBD children

目的 为探查外周血有白细胞坏死的大骨节病（ＫＢＤ）病儿其血浆内有无ＫＢＤ的致病因子，特进行本实验，方法 选干骺端损害Ｘ线阳性征且有外周血细胞坏死的ＫＢＤ病儿血浆作用于培养（兔）软骨细胞以判定血浆内有无引起软骨细胞病变效应的致病因子。结果 实验组部分细胞的核染色质颗粒间出现淡紫红色小点状物、伴点状物周围染色质溶解，直至整个胞核转化为青灰均质的大团块；更后，团块内出现嗜酸性斑块、团块外出现环状体。电镜下见实验组细胞核内众多染色质溶解小灶、各小灶内有直径７５～８３ｎｍ的病毒核衣壳，呈外形六角、核心中空。结论 以ＫＢＤ病儿血浆诱发出的培养软骨细胞的光镜下病变与ＫＢＤ尸骨软骨细胞的Ｂ型凝固性坏死完全相同，实验复制出ＫＢＤ的培养软骨细胞病变模型；病变的原因是来自ＫＢＤ病儿血浆、继之出现于实验组病变细胞核内的某病毒；ＫＢＤ病儿血浆内的病毒可能就是ＫＢＤ的致病因子。

Objective To probe whether there was a causal factor for Kaschin-Beck diease( KBD) in blood plasmas of KBD children with the necrosis of white blood cells in peripheral bloods，we set up the damage experiment. Methods The plasmas of KBD children with the positive X-ray signs ci metaphyseal lesions and the necrosis of white blood cells in peripheral bloods were added to cultural rabbit chondrocytes，so that the judgement whether there was a causal factor for KBD in the plasmas could he done on the basis of a cytopathic effects in the cultural chondrocytes. Re- sults In the experimental group，some light purple-red dotted substances developed among the chromatin granules asso- ciated with chromatinolysis round the dotted substances in nuclei of partial chondrocytes，until these nuclei transformed in- to a light grey and homogeneous masses，later，some acidophilic spots developed within the mass and a ring outside the mass. By transmission electron microscope，very much small foci of chromatinolysis developed within the cell nuclei of the- experimental group，and there was a viral nucleocapsid with 75~83nm in diameter that outline was hexagon and center was empty in every focus. Conclusions The lesions observed by optical microscopy in the cultural chondrocytes induced by optical microscopy in the cultural chondrocyed by adding the plasmas of KBD children were fully coincided with the le- sions of the type B coagulation necrosis of chondrocytes in autopsy samples with KBD. A pathological model ci the cultural chondrocyte for KBD had been reproduced in this experiment The cause of Pathological changes was the virus which was from the plasmas of KBD children and secondarily developed in the nuclei of lesion cells in the experimental group. The vinis in the plasmas of KBD chilren might be the causal factor for KBD.
