摘要 目的 大鼠慢性染镉后给予GSH、Vit C、DMPS ,探讨这 3种物质对镉肾毒性的影响。方法 将SD大鼠 5 0只 ,随机分成 5组。第 1组为对照组 ,第 2组为单纯染镉组 ,第 3~ 5组为染镉后再投与GSH、Vit C、DMPS组。先给第2~ 5组大鼠皮下注射 5 μmol/kg的氯化镉溶液 ,每天 1次 ,每周 5次 ,共 11周 ,对照组在相应时间内皮下注射 0 9%氯化钠。以上各组均在最后一次注射后收集尿样。第 12周开始 ,第 2组动物腹腔注射 0 9%氯化钠 ,第 3组动物腹腔注射 1mmol/kg的GSH ,第 4组动物腹腔注射 4mmol/kg的Vit C ,第 5组动物腹腔注射 0 2mmol/kg的DMPS ,对照组在相应时间内腹腔注射 0 9%氯化钠。每周 3次 ,连续 2周。最后一次注射后 2 4h再次收集尿样 ,对 2次收集的尿样测定其NAG活性、蛋白含量及镉含量。结果 染镉后 ,尿NAG活性、尿蛋白含量及尿镉含量与对照组比明显增高 ,投与GSH、Vit C、DMPS后 ,与投与前和单纯染镉组比较尿蛋白含量和尿NAG活性均明显下降 ;GSH和Vit C投与组及单纯染镉组尿镉含量明显下降 ,而DMPS投与组尿镉含量无明显变化。结论 慢性染镉 11周引起了肾脏损伤 ,给予大鼠GSH、Vit C、DMPS对镉引起的慢性肾损伤的恢复有一定促进作用。 Objective GSH,Vit-C and DMPS were given to rats to study the effects on the renal toxicity induced by chronic exposure to cadmium.Methods Fifty SD rats were divided into five groups at random.Four groups rats were given subcutaneous injections of 5μmol/kg CdCl\-2 and another group rats were injected with 0\^9% NaCl for eleven weeks.The urine sample were collected after the last injection.From the twelfth week,the sceond group rats were intraperitonealy(ip) injected with 0\^9% NaCl,the third group rats were ip injected with 1mmol/kg GSH,the fourth group rats were ip injected with 4mmol/kg Vit-C and the fifth group rats were ip injected with 0\^2mmol/kg DMPS.After two weeks,urinary samples were collected again.NAG activities,protein and cadmium contents were determined in the urinary samples.Results Urinary NAG activities,contents of urinary protein and cadmium in the Cd-administered groups were increased significantly,and they were different from the control group.After the administration of GSH,Vit-C and DMPS,urinary NAG activities and the contents of urinary protein was decreased significantly as compared with that before the administration of the three compounds,and with that of the animals given cadmium alone.The urinary cadmium contents were decreased significantly except the animals that were administered with DMPS.Conclusion Cadmium administration resulted in the renal injury,and GSH,Vit-C and DMPS played the antagonistic effects on the renal toxicity of cadmium.
