摘要
探讨气虚证模型大鼠A区基因表达谱特征及其对治疗的反应。采用药物加疲劳法制造Wistar大鼠气虚模型 ,AtlasTM反义DNA(cDNA)阵列法检测造模动物鼻咽上皮细胞基因组A区基因表达谱。结果 :模型组动物表现细胞核转录因子亚单位基因c -jun和c-kit原癌基因表达下调 ,前胸腺素α和钙结合蛋白 2基因表达上调。治疗 1周后 ,表达上调的 2基因活性恢复正常 ,但表达下调的 2基因活性仍维持低表达状态。结论 :单纯气虚证动物出现原癌基因和应激反应相关基因表达活性异常 ,其变化趋势和病理生理意义在于尽力维持机体内环境的稳定 ,即阴阳平衡。
To investigate the features of gene expression profile on A area of Atlas TM cDNA array and their response to treatment among rats with qi deficient syndrome.Qi deficient syndrome was induced in rats with the method of herbal decoction perfused gastrically and supplemented by forced swimming. The expressive features of genes on A area were determined in nasopharyngeal tissues of tested rats on Atlas TM cDNA expression array.Results: The expressive activities of c jun and c kit proto oncogenes were downwards regulated,and those of prothymosin α and calcium binding protein genes were upwards regulated among the rats of model group,while c jun and c kit were the only genes still remaining in an abnormal status of activity,which were downwards regulated,among the rats of the treatment group.Conclusion:The features of gene expression profile of rats with qi deficient syndrome were the abnormal expression activities of proto oncogenes and those genes related with stress response.The implications of this pathological process lie in the maintenance of the balance of yin and yang within the diseased body to a most favorable degree.
