中国人CTLA-4第一外显子的基因序列分析

The gene sequence of cytotoxic T-lymphocyte antigen 4 exon 1 in Chinese peripheral blood

目的 研究正常人外周血细胞毒性T淋巴细胞抗原 4(CTLA 4)的基因序列。方法 采用PCR扩增正常人外周血 ,单个核细胞中CTLA 4第一外显子的基因片段并以Model377测序仪进行序列分析。结果 得到 16 2bp的PCR产物 ,与预计结果相符 ,基因序列与GeneBank中序列相同。结论 正常中国人CTLA 4第一外显子基因序列与国外报道一致。本方法建立为研究CTLA 4基因多态性与自身免疫性疾病关系奠定基础。

Objective To investigate the normal Chinese gene sequence of cytotoxic T-lymphocyte antigen 4(CTLA-4)from peripheral blood.Methods CTLA-4 exon 1 was amplified by PCR and sequenced by Model 377 sequencer.Results The sequence of 162bp PCR product was obtained and the sequence was in highly harmony with that from Gene Bank.ConclusionThe normal Chinese gene sequence of CTLA-4 exon 1 accord with those reported from other countries.This method is simple and reliable for studying the relationship between gene polymorphism of CTLA-4 and some autoimmune diseases.
