摘要
目的 研究失血致低血容量性休克大鼠心肺组织G蛋白的变化。方法 颈总动脉放血 ,使平均动脉压降至 5 .9kPa ,稳定 1h ,致大鼠中度失血致低血容量性休克模型 ,6h后用Westernblot法测定心肺组织Gsα、Giα、Gqα、Goα的含量。 结果 休克后心脏膜组织Gsα两条带较对照组显著降低。Giα和Gqα带较对照组则显著升高 ,Goα降低 (P <0 .0 5 )。肺膜组织Gsα两条带较正常对照组显著降低。Giα和Gqα较正常对照组也下降 (P <0 .0 5 )。结论 大鼠心肺组织G蛋白的变化可能参与了失血致低血容量性休克时信号转导系统功能障碍的发生。
Objctive To study the alteration of the content of G-proteins α subunits of rats in shock.Methods Hemorrhagic shock was performed in Wistar rats by carotid artery bleeding to maintain the mean arterial pressure(MAP) at 5.98kPa(45mmHg) for one hour.Fourteen Wistar rats were randomly divided into two groups.Group 1(NS,n=7),as control;Group 2(HS n=7),shock control.Determining the blood gases and mean arterial pressure,then,the content of Gsα,Giα,Gqα,Goα in the tissues of the ventricles and lung membranes tissue isolated from rats were measured by Western Blot analysis at 6 hours after shock.Results In the ventricles,the content of two Gsα bands with molecular weights of 52KD and 45KD-(84.2±5.8)% and (84.6 ±4.2)%;One Goα band with molecular weights of 39KD-(90.6±4.7)% in the group HS was significantly lower than in the group NS(100%),P<0.05,<0.01and P<0.05.The content of one Giα band and Gqα band with molecular weights of 41kD and 42kD-(159.4±7.2)% and (130.3±20.9)% in the group HS was significantly higher than in the group NS,pP<0.01 and P>0.05.In the tissues of lung ,the change of the content of two Gsα band is as same as in the ventricle,P<0.05.The content of one Giα band was lower in the group HS(P<0.05).Conclusion The change of the content of G-proteins of the ventricles and lung tissue of the rats,may involve in the development of the hemorrhagic shock.
