摘要
目的 探讨脉冲Nd∶YAG激光对培养肥厚性瘢痕成纤维细胞胶原合成的选择性抑制作用。方法 以不同能量密度(5 0 0J/cm2 、10 0 0J/cm2 、15 0 0J/cm2 和 2 0 0 0J/cm2 )脉冲Nd∶YAG激光 (波长 10 6 4nm ,脉宽 15 0 μs)照射培养肥厚性瘢痕和正常皮肤成纤维细胞 ,照射后 2 4h分别用 3H 脯氨酸掺入法和斑点杂交法检测成纤维细胞胶原合成和Ⅰ型前胶原基因表达水平。结果 肥厚性瘢痕与正常皮肤成纤维细胞胶原合成、Ⅰ型前胶原mRNA水平在 5 0 0J/cm2 照射后无变化 ,15 0 0J/cm2 、2 0 0 0J/cm2 照射后显著降低 (P <0 .0 0 1) ;10 0 0J/cm2 照射后 ,肥厚性瘢痕成纤维细胞胶原合成与Ⅰ型前胶原基因表达水平显著下降 (P <0 .0 0 1) ,而正常皮肤成纤维细胞胶原合成和Ⅰ型前胶原基因表达水平却无变化 (P >0 .0 5 )。结论 一定能量密度 (10 0 0J/cm2 )脉冲Nd∶YAG激光对体外培养肥厚性瘢痕成纤维细胞胶原合成具有选择性抑制作用。
Objective In order to explore the selectively inhibitor effects of pulsed Nd:YAG laser irradiation on collagen production of cultured fibroblasts of hypertrophic scar(HS).Methods Cultured fibroblasts derived from HS and normal human skin were irradiated with a pulsed Nd:YAG laser(wavelength 1064nm,pulse width 150μs)at various energy density levels (500J/cm 2,1000J/cm 2,1500J/cm 2and 2000J/cm 2).At 24 hours after laser irradiation,collagen production of fibroblasts was measured by the incorporation of proline.The expression of proαl(Ⅰ)procolagen mRNA was investigated by blot hybridization techniques.Results Collagen synthesis and type I procollagen mRNA level in HS and normal dermal fibroblasts remained unchanged at energy level of 500J/cm 2,and were significantly decreased at energy densities of 1500 and 200J/cm 2(P<0.001).Collagen production and type I procollagen mRNA level in HS fibroblasts were markedly reduced at energy density of 1000J/cm 2 (P<0.001),the dose that did not affect collagen synthesis and type I procollagen mRNA level in normal dermal fibroblasts(P<0.05).Conclusion Pulsed Nd:YAG laser at energy density of 1000J/cm 2 can selectively suppress collagen synthesis of cultured fibroblasts in HS.
