Inhibitory effect of interleukin-13 on LPS-induced expression of interleukin-1βin mesangial cells by down-regulated activation of nuclear factor-κB

摘要 目的 研究白细胞介素13(IL-13)对脂多糖(LPS)引起的肾小球系膜细胞核因子-κB(NF-κB)活化及白细胞介素1β(IL-1β)表达的影响,以探讨其抗炎作用的机制。方法 以体外培养的大鼠肾小球系膜细胞为靶细胞,用凝胶电泳迁移率试验(EMSA)测定NF-κB活性;用酶联免疫吸附法(ELISA)及逆转录聚合酶链反应(RT-PCR)观察系膜细胞 IL-1β蛋白及基因表达。结果 LPS可诱导系膜细胞NF-κB活化及IL-1β表达,这作用能被NF-κB抑制剂吡咯二硫氢基甲醋酯(PDTC)抑制。IL-13亦有此抑制作用。结论IL-13对于系膜细胞炎症反应的抑制作用可通过抑制NF-κB活化而实现。 Objective To examine the effect of interleukin-13(IL-13) on expression of interleukin-1β-(IL-1β) and activation of nuclear factorκB(NF-κB) induced by LPS in mesangial cells and to explore the anti-inflammatory mechanism of IL-13. Methods The protein synthesis of IL-1β in mesangial cells was measured by ELISA. The expression of IL-1β mRNA in mesangial cells was examined by RT-PCR. The activation of NF-κB in mesangial cells was determined by electrophoresis mobility shift assay(EMSA) . Results LPS induced activation of NF-κB and the expression of IL-1β in both protein and mRNA levels in mesangial cells. NF-κB inhibitor PDTC as well as IL-13 could inhibit the above effect of LPS. Conclusion Inhibitory effect of IL-l3 on IL-1β expression induced by LPS is mediated by, at least in part, down-regulation of NF-κB activation.
