摘要
目的 :探讨肾上腺髓质素 (adrenomedullin ,AdM )对同型半胱氨酸 (homocysteine ,HCY)诱导的气道平滑肌细胞增殖的影响和HCY对肺组织合成分泌AdM的影响。方法 :在培养的大鼠气道平滑肌细胞上 ,测定3 H -TdR参入 ;用放免法测定肺组织和孵育液中AdM含量。结果 :0 .5mmol·L 1HCY刺激气道平滑肌细胞 (airwaysmoothmusclecells ,ASMC)增殖 ,AdM (10 9,10 8,10 7mol·L 1)呈浓度依赖方式抑制HCY诱导的ASMC增殖 ;HCY(0 .1,0 .5 ,1.0mmol·L 1)以浓度依赖方式诱导肺组织合成和分泌AdM ,超氧化物歧化酶 (superoxidedismutase ,SOD)可抑制其作用。结论 :在肺HCY和AdM呈相互作用关系 ,HCY调节AdM的生成 ,而AdM影响HCY的生物学效应。
Objective:To study the effect of Adrenomedullin (AdM) on the Homocysteine(HCY) induced proliferation of airway smooth muscle cells and the effect of HCY on the AdM synthesis and secretion in lung tissue.Methods:The 3 H TdR incorporation was measured in cultured airway smooth muscle cells(ASMC),and the content of AdM in lung tissue and incubative fluid were measured by a radioimmunoassay. Results:HCY (0.5 mmol·L -1 ) induced the proliferation of ASMC.AdM(10 -9 ,10 -8 ,10 -7 mol·L -1 ) inhibited the HCY induced proliferation of airway smooth muscle cells in a concentration dependent manner . HCY induced the AdM synthesis and secretion in lung tissue in a concentration dependent manner,and this effect was inhibited by superoxide dismutase (SOD). Conclusion: HCY and AdM influence each other in the lung, HCY regulates the production of AdM,which,in turn, inhibits the biological effect of HCY.
