摘要
以豚鼠离体气管平滑肌为实验材料,采用生物检测法,观察野罂粟总生物碱和Ⅰ、Ⅴ、Ⅵ号单体对豚鼠离体气管平滑肌白三烯D4所致收缩的影响,结果表明,野罂粟总生物碱和Ⅴ号单体能竞争性拮抗白三烯D4所致收缩,IC50分别为47.6μg.ml-1和19.7μmol.L-1;以大鼠胸腔多形核白细胞为实验材料,采用反相高效液相层析紫外检测法,检测药物对白三烯B4生物合成的影响,结果表明,野罂粟总生物碱和Ⅵ号体对LTB4的合成具有抑制作用,此作用与抑制磷脂酶A2的活性无关,很可能是通过抑制5-脂氧酶的活性实现的。
This article reports the action of to tal alkaloid of papaver nudicaule(TAPN)and I,V,VI monomers on LTD4and LTB4.Experiments by bioassay were p erformed that TAPN and V competitively antagonized LTD4the constrictiv e effect of isolated tracheal smooth muscle o f guinea pigs induced by LTD4,IC50we re 47.26μg.ml -1,respectively.Mean-while,TAPN and VI inhibited biosynt hesis of LTB4in rat pleural polymorp h nuclear leukocytes,IC50were62.99μg.ml -1and 42.3μmol.L -1respectively,which had no r elationship with inhibition of acti vity of phospholipase A2.These results indicate TAPN and i ts monomers have antagonisitical effect on leukotrienes.
