摘要
以PvSR6cDNA编码一种菜豆DnaJ like蛋白 ,并利用PCR的方法克隆出基因组部分序列 .核苷酸序列分析表明PvSR6基因在这段编码区内无内含子 ;Southernblot分析表明菜豆基因组中存在多个PvSR6基因拷贝 ;Northernblot分析结果表明PvSR6是组成型表达蛋白 ,重金属Hg、Cd和As ,过量的Cu和Zn及受伤、高温、病毒侵染和水杨酸等均能强烈地诱导其基因在叶片中的表达 ,表明DnaJ
PvSR6 ( Phaseolus vulgaris Stress Related)cDNA encodes a bean ( Phaseolus vulgaris L.)DnaJ like. Sequence analysis of partial genomic DNA showed no intron in its coding region. Southern blot showed that more than one PvSR6 gene copy was existed in bean genome. Northern blot revealed that PvSR6 gene was constitutively expressed in leaf tissue, however, the mRNA level was increased by heavy metals (Hg, Cd and As),excess Cu and Zn ions, and other environment stresses, such as wounded, virus infection, elevated temperature and salicylic acid. It suggested that PvSR6 might be involved in the process of plant resistance to various stresses.
