细胞因子和免疫抑制剂对人胚胎心肌细胞HLA抗原表达的影响

摘要 我们的实验表明,正常人胚胎心肌细胞(FHC)检测不出人类白细胞抗原(HLA)。激活与未激活的正常人外周血淋巴细胞均能诱导FHC表达HLAⅠ类和Ⅱ类抗原。α干扰素(IFN-α)、白细胞介素1、3、4(IL-1、3、4)以及α肿瘤坏死因子(TNF-α),都能诱导FHC表达HLAⅠ类而不是Ⅱ类抗原。IFN-γ是Ⅰ娄和Ⅱ类抗原的诱导剂。值得注意的是,我们发现IFN-α和TNF-α联合应用时,不仅对Ⅰ类抗原的诱导起增强作用,而且能协同Ⅱ类抗原的表达。环孢霉素A(CsA)和FK-506免疫抑制剂均能抑制Ⅱ类及Ⅰ类抗原的表达。 Our data indicated that human fetal heart cells (FHC) fail to express either HLA class I or class II antigens. Mixed lymphocyte culture or oonoanavalin A activated lymphocytes and unstimulated normal human peripheral blood lymphocytes can all induce expression of class I and class II HLA antigens on FHC. Alpha-interferon (IFN-α), interleukin 1(IL-1), IL-3, IL-4, and alpha-tumor necrosis factor (TNF-α), can all induce expression of class I but not class II HLA antigens on FHC. It is very interesting that synergistio interaction of IFN-α and TNF-α induces expression of class I and class II HLA antigens on cardiac myooytes. Immunosuppressive agenbs both cyclosporin A and FK-506 strongly inhibit expression of class I and class II HLA antigens on fetal cardiac myocytes.
