摘要
用Con A活化的小鼠脾脏细胞免疫同品系动物(BALB/c鼠),取免疫鼠的脾细胞与小鼠骨髓瘤细胞(SP2/0)融合,选择了一株有分泌抗活化小鼠T细胞抗原抗体的杂交瘤细胞,经有限稀释法连续4次克隆化。采用ELISA法分析,初步证实该杂交瘤细胞所分泌的单克隆抗体(McAb)与Con A 活化的小鼠脾细胞的结合反应明显高于与未经活化的其他细胞(胸腺细胞、肠系膜淋巴结细胞、脾细胞)的反应。用琼脂双扩散法证明该MeAb属于小鼠IgG类。
We succeeded in selecting a hybridoma cell line secreting antibodies against activated mouse T lymphocyte antigen by means of fusing between mouse myeloma cells (SP2/0) with spleen cells from BALB/o mice immunized with Oon A aotivaed syngenio spleen cells.The hybridoma cell line wes recloned 4 times successively by a limiting dilution method.Preliminary analysis by ELISA showed that the binding reaction of this monoclonal antibody to activated mouse spleen cells was significantly stronger than with other non-stimulated cells (thymocytes,mesentery lymph node cells and spleen cells).The monoclonal antibody is proved by double immunodiffusion method to be of the mouse IgG class.
