摘要
我们从人脑和肝组织中分离获得的神经节苷脂单组分GM_1和GM_3对兔红细胞膜钠泵活性影响进行了研究。GM_1的组成中比GM_3多一个半乳糖-N-乙酰半乳糖残基。实验表明:GM_1对兔红细胞膜钠泵有激活作用,而GM_3则为非竞争性抑制作用,V_m发生改变而K_m不变。提示外源性GM_1和GM_3的掺入,对钠泵的激活和抑制作用,可能通过诱导细胞膜微环境的改变,或直接结合于膜上的特殊位点,引起钠泵构象的改变而影响其活性。
Pare gangliosides GM1 and GM3 were isolated from human brain and liver tissues.Structurally,GM1 contains more Gal-Gal(NAc)-residue than GM3.The characteristics of GM1 and GM3 effects on the rabbit red cell sodium pump were studied.The results indicate that GM1 activates the sodium pump,but GM3 is a non-competitive inhibitor of the sodium pump in rabbit red cells.It was suggested that the mechanism of incorporated gangliosides may modify the mioroenvironment of cell membrane,or combine directly to specific sites on the cell membrane to modulate the conformation of the sodium pump and then influence its activity.
