摘要
本文报道全反式维甲酸(ATRA)对高分化胃腺癌 MKN-28细胞似具有生长抑制及诱导分化作用,使胃癌细胞内 DNA 合成减少,并阻止 G1/0期细胞进入 S 期,同时降低 G2/M 期细胞的比例。对 MKN-28细胞恶性程度检查的结果表明,ATRA 可降低 MKN-28细胞在双层软琼脂中的克隆形成,其抑制率为60.3±6.4%;同时可以推迟 MKN-28细胞在裸鼠体内移植瘤倍增生长的时间及减慢其生长速度,但不能阻止移植瘤形成。形态学观察表明,ATRA 不能改变移植瘤HE 染色及 AB/PAS 染色的特性。
This study demonstrates the proliferation inhibitory and differentiation inducing effectsof all-trans-retinoic acid(ATRA)on well-differentiated gastric cancer cell line MKN28.The results showed that the proliferation of MKN28 cell line was inhibited by ATRA(inhibitory rate 46.7%),while ~3H-thymidine incorporation of the cell line was also inhibited,and the cell cycle of the cell line was changed by ATRA.In the ATRA-treated MKN28cell line,ability of clony formation in soft agarouse was impaired(inhibitory rate 60.4±6.4%),a part of shuttle-like cells appeared in vitro,the growth rate and weight of transplanted gastric cancer in athymic nude mouse were reduced,but its tumorigenicity wasnot influenced and ultrastructure of the transplanted tumors was not different from thecontrol group.
