维甲酸与1,25(OH)_2D_3对白血病细胞的联合促分化效应 被引量:3

摘要 本文报道了13-顺式维甲酸(RA)和[1,25-二羟维生素 D_3 1,25(OH)_2D_3]合用对两种白血病细胞系的体外实验结果。在10^(-7)mol/L 浓度下,应用 ~3H-TdR 掺入、NBT 还原试验、流式细胞分析等技术,测得:RA 和1,25(OH)_2D_3合用后可进一步抑制细胞的增殖;在诱导分化作用方面,二者对人原始巨核白血病细胞株(HIMeg)有相加作用,而对人早幼粒细胞白血病细胞系(HL-60)则有一定协同作用。上述结果提示可将 RA 联合1,25(OH)_2D_3试用于治疗白血病;推测二者在体内巨核系和粒单系造血细胞分化中发挥着相加或协同的促进效应。 The action of 13-cis-retinoic acid(RA)in combination with 1,25-dihydroxyvitaminD_3(D_3)on two kinds of leukemia cell lines in vitro is reported.At the concentrationof 10^(-7) mol/L,RA and D_3 could inhibit the cell proliferation and they have additiveeffect on the human promyelocytic leukemia cell line(HL-60),and also have synergiceffect on the human megakaryoblastic leukemia cell line(HIMeg)in inducing celldifferentiation.The results suggest that RA in combination with D_3 may be used forthe treatment of acute megakaryoblastic leukemia and acute promyelocytic leukemia.It ispresumed that they had additive or synergic action in the differentiation of hemopoieticcells in vivo.
