摘要
本文主要研究中药槲寄生、川芎嗪对体外原代人脐静脉内皮细胞形态和功能的影响。槲寄生浓度为200μg/6×10<sup>4</sup>细胞及100μg/6×10<sup>4</sup>细胞时,不同程度地损伤细胞形态,抑制内皮细胞合成释放 PGI<sub>2</sub>,但经凝血酶激发后的内皮细胞合成释放前列环素 I<sub>2</sub>(PGI2)功能未受影响。此外,200μg/6×10<sup>4</sup>细胞浓度的槲寄生还抑制凝血酶激发前后内皮细胞合成释放 Von Willibrand 因子(vWF)。川芎嗪高、中、低三种浓度均未影响细胞形态和细胞合成释放 PGI<sub>2</sub>及 vWF 的功能,鉴于其能抑制血小板聚集及血栓素 A<sub>2</sub>(TXA<sub>2</sub>)释放,提示川芎嗪是改善 PGI<sub>2</sub>—TXA<sub>2</sub>平衡的理想药物。
The effects of Viscum coloratum and tetramethylpyrazine on morphology and func-tions of cultured endothelial cells originated from human umbilical veins were studied.Thecells were morphologically damaged in various degrees and their functions on producing andreleasing prostaglandin I_2(PGI_2)were inhibited when the Viscum coloratura concentrationswere 200 ug/6×10~4 cells and 100 ug/6×10~4 cells.But after the endothelial cells werepretreated with thrombin,neither the synthesis nor the release of PGI_2 would be effected.Furthermore,Viscum coloratum 200 ug/6×10~4 cells could inhibit the function of enothe-lial cells in producing and releasing Von Willibrand Factor(VWF)before or after acti-vating by thrombin.While tetramethylpyrazine in any concentration had no effects on cellmorphology and functions of producing or releasing PGI_2 and VWP.Since tetramethylpyrazine could both inhibit platelet aggregation and thromboxine A_2(TxA_2)synthesis,it ispossibly an ideal agent for ameliorating the PGI_2-TxA_2 equilibrium.
