大鼠肺纤维化模型肺泡上皮钙粘素的表达 被引量:15

摘要 目的 :探讨钙粘素 (cadherin)在肺纤维化发病机制中的作用。方法 :SD大鼠 48只随机分为实验组和对照组 ,每组 2 4只。实验组气管内滴入博莱霉素建立肺纤维化模型 ,使用SABC免疫组化方法检测实验组滴药后第1,2和 4周及对照组肺泡上皮钙粘素的表达 ,利用图像分析技术测定阳性表达面积。结果 :对照组钙粘素表达较低 ,阳性表达面积为 (96 8.5± 138.11) μm2 。给予博莱霉素后表达上调 ,实验组第 1,2和 4周阳性表达面积分别为 (20 80 .9± 42 9.0 9) μm2 、(130 9.3± 35 7.0 0 ) μm2 和 (1138.4± 337.2 1) μm2 。实验组阳性表达面积与肺泡炎积分呈正相关(r=0 .786 ,P <0 .0 1)。结论 :钙粘素参与肺纤维化的发病过程 。 Abstract: To investigate the role of cadherin in pathogenesis of pulmonary fibrosis. Methods: The expression of cadherin of alveolar epithelium cells was evaluated by immunocytochemistry and image analysis methods on rats with pulmonary fibrosis induced by bleomycin. Results: The positive expression area of cadherin in alveolar epithelium cells was low in control group, only (958.5±138.11)μm 2. There was an increase in the experimental group. The positive expression area in the first week, the second week and the fouth week group was (2 080.9±429.09) μm 2,(1 309.3± 357.00 ) μm 2and (1 138.4±337.21) μm 2. There was positive correlation between the positive expression area and the score of alveolitis in the experimental group (r=0.786,P<0.01). Conclusions: Cadherin has close relationship with the pathogenesis of pulmonary fibrosis, expecially at the stage of alveolitis.
