摘要
目的 :探讨不同剂量、不同溶剂的基因重组技术生产的组织型纤溶酶原激活物 (r tPA) ,对于不同脱离时间的视网膜毒性作用。方法 :采用经外路显微手术方法 ,将Healon注入脉络膜视网膜下腔 ,制作出 30只脉络膜视网膜脱离兔眼模型 ;将不同剂量、不同溶剂的r tPA注入不同网脱天数的玻璃体腔内 ;用ERG、光学显微镜、电子显微镜等方法测定实验效应 ;用正交设计方法分析实验结果。结果 :脱离的视网膜对r tPA的毒性耐量明显降低 ,且脱离的时间越长 ,视网膜对r tPA耐量越低 (P <0 .0 1) ;不同的溶剂亦对这种耐受性有较明显的影响 (P <0 .0 5 )。结论 :脱离状态的视网膜对r tPA的毒性耐量降低 。
Abstract: To investigate the toxicity of r tPA to detached retinal under the condition of different doesage, different solution and different retinal detachment time. Methods: Injecting Healon into choroids and retinal subcavity by outer way method, which establishing 30 case rabbit retinal detachment models. Then injecting r tPA of different doesage, different solution into vitreous cavity which detachment time of retinal was different; measure the experimental effect by ERG, lightmicroscope, electron microscope; analyse the results of experiment by orthogonal design. Results: (1) The r tPA tolerance of detached retinal decreasing obviously than undetached retinal (P<0.01), and the longer the retinal detached, the lower the tolerance is. (2) Different solution can also influence the tolerance obvious (P<0.05). Conclusion: The tolerance to r tPA of detached retinal decreasing obvious which should be took care in clinical work.
