荧光脱氧葡萄糖显像的初步试验研究 被引量:1

摘要 目的 :研究利用脱氧葡萄糖的荧光标记类似物 6 - [N - (7-硝基苯基 - 2 -氧杂 - 1,3-重氮盐 - 4-基 )氨基 ]- 2 -脱氧葡萄糖 (2 -NBDG)进行荧光显像的可行性。方法 :本试验利用了 3只雄性小鼠。鼠 1饥饿 16h后在室内光照条件下由尾静脉注入 0 .0 2 6mg/ 0 .1ml的 2 -NBDG ,然后该小鼠保持室内光照直至处死。鼠 2正常进食 ,在完全黑暗的环境下由尾静脉注入 0 .0 2 6mg/ 0 .1ml的 2 -NBDG ,然后该小鼠保存在黑暗环境下直至处死。鼠 3在室内光照条件下由尾静脉注入 0 .1ml的生理盐水 ,然后该小鼠保存在室内光照环境直至处死。 3只小鼠均在注射后 30min处死 ,其中鼠 2是在黑暗环境下处死 ,鼠 1和鼠 3在室内光线下处死。处死后分离 3只小鼠的大脑并进行水平方向切片 ,层厚 10 μm。我们选用每只小鼠视皮质部位的 7个切片 ,在一个大视野的CCD系统下进行荧光显像。结果 :显像结果表明 ,在接受 2 -NBDG注射的 2只鼠 (鼠 1和鼠 2 )的大脑内可以见到明显的荧光。而未接受 2 -NBDG注射的鼠 3大脑内则未见任何荧光。鼠 1双侧视皮质对应部位荧光强度与周围皮质无明显差别 ,而鼠 2双侧视皮质对应部位荧光强度则略低于周围皮质。对鼠 1和鼠 2大脑荧光图像的定量分析表明 ,鼠 1视皮质部位的平均荧光强度明显高于鼠 2 Objective: The purpose of this study is to demonstrate the possibility of detecting the fluorescence of the glucose fluorescent analogue 2-NBDG. Methods: Three male mice were used in this experiment. Mouse 1 was fasted over 16 hours, injected 0.026 mg 2-NBDG through tail vein in room light environment and keep in light environment until sacrificed. Mouse 2 was fed, injected 0.026 mg 2-NBDG in completely dark environment and keep in this environment until sacrificed. Mouse 3 was injected 0.1 ml saline in light environment and keep in light environment. Both mice was sacrificed 30 minutes after injection. Mouse 2 was sacrificed in dark environment, the others were sacrificed in light environment. The brain of the three mice were separated and sectioned horizontally. We selected 7 visual cortex slices of each mouse and imaged it under a large field of view CCD system. Results: The images showed that there was significant fluorescence in the rats receiving 2-NBDG (mice 1 and mice 2). In the mouse that not receiving 2-NBDG, the image was completely dark. The images of mouse 2 showed that the right and left visual cortex area was slightly darker than the surrounding cortex. Quantitative analysis showed that the average gray scale of visual cortex of mouse 1 was significantly greater than mouse 2 ( P <0.01). Conclusion: CCD system can successfully detect the distribution of 2-NBDG and the brain glucose metabolism level can be reflected by this method.
