疏水层析介质苯基-琼脂糖凝胶合成与吸附性能的研究

摘要 以琼脂糖凝胶为基质 ,与 2 ,3-环氧丙烷苯基醚反应后 ,制得疏水层析介质苯基 -琼脂糖凝胶。考察了它的物化性能及对多种蛋白、基因产品的吸附与分离性能 ,结果表明其吸附性能、分离纯化性能、化学稳定性均与国外同类产品相近。对链激酶的纯化收率大于 95 % ;人血 B因子的纯化倍数为 2 6 ,收率大于 95 %。 The phenyl-agarose gel was prepared by reacting 2,3-glycidol phenyl ether with Sepharose CL-4B and its physical and chemical properties were determined. The adsorption and purification properties of the gel were investigated by adsorptio n a nd elution of various proteins. The results showed that the adsorption capacity and physical and chemical stability of the phenyl-agarose gel were comparable to imported analogous product. The purifying multiple of human blood B factor (HBBF ) were 26 times and the recovery of HBBF and streptokinase were greater than 9 5% respectively.
