摘要
目的:研究胎盘型谷胱甘肽转移酶(GST-π)在乳腺癌及其腋窝淋巴结组织中的表达情况与乳癌发生和临床耐药的相关性。方法:采用免疫组化及RT-PCR法分析临床乳癌标本及肢窝清扫淋巴结组织GST-π蛋白和mRNA的表达。结果:在正常乳腺组织、乳癌组织、HE阳性淋巴结、HE阴性淋巴结中GST-π蛋白阳性表达率分别为0,60%,77%,43%;GST-π mRNA阳性表达率分别为:0,88%,7l%,39% 。结论:在肿瘤启动和促进阶段GST-π表达增高,是一典型癌前病变标志酶;HE阴性淋巴结中出现GST-π阳性表达,提示预后不好,且在耐药反应性上淋巴结灵敏性强于乳癌组织;GST-π基因表达的调控可能主要发生在RNA和(或)转录前水平。
Abstract Objective:To study GST-π expression in breast cancer and axillary lymphnodes.Methods: The expression of GST-Tr proteinand mRNA was detected by means of immunohistochemistry and RT- PCR. Results: In normal breast tissue, cancer tissue , HE positive lymph-nodes and HE negative lymphnodes , GST-π protein expression rate is 0,60% ,77% ,43%; GST-π mRNA expression rate is 0,88% ,71%and 39% , respectively. Conclusion; GST-πgene whose expression is upregulated as tumor occurs and develops can be used as a tumor biomar-ker; GST-π expression in HE negative lymphnodes suggests the poor prognosis; drug-resistance response is more sensitive in lymphnode than inprimary cancer. The expression of GST-π gene may be regulated by RNA or before RNA transcription.
