毛细管气相色谱法测定人血红细胞膜上中性糖、氨基糖及唾液酸的含量

Determination of Neutral Sugars,Hexosamins and Silaic Acids of Erythroc yte Membrane by Capillary Gas Chromatography(CGC)

用肌醇作内标物 ,对人血红细胞膜上中性糖、氨基糖及唾液酸的含量进行毛细管气相色谱分离、分析。通过低温差速离心从人血中获取纯净的红细胞膜。用酸水解多糖链后经离子交换净化 ,再用醋酐、吡啶加热回流乙酰化。各组分对不同加入量的回收率在 95 .96 %～ 98.2 1%之间。重复性试验中各组分相对平均偏差在 3.5 2 %～ 7.0 6 %之间。方法简便可靠。

Neutral,amino sugars and silaic acids in erythrocyte m embrane were determined by CGC using inositol as internal standard(i.s.). By me ans of low temperature differential-speed centrifuge, pure erythrocyte membrane w as obtained from blood. The glycosides in membrane proteins were extracted by us ing acid and cleared by using ion exchange column. The monosaccharides were acet ylated with acetic anhydride and pyridine. Recovery obtained by adding differe nt amo unts of monosaccharides was 95.96%～98.21%. CV of determination was 3.52%～7.06 %.The method was proved to be simple and reliable.
