摘要
目的 :研究脓毒症时大鼠急性期反应以及急性期反应产物合成调控与内毒素 (L PS)水平的关系。方法 :雄性 Wistar大鼠 40只 ,腹腔注射内毒素复制脓毒症模型 ,随机分为正常组 ,内毒素攻击后 2、6、12、2 4小时组。应用改良过氯酸预处理血浆 ,基质显色法鲎试剂 (L AL )和免疫比浊法测定血浆 L PS和 C反应蛋白 (CRP)浓度。结果 :L PS和 CRP在内毒素攻击后均显著升高 ,但 L PS在内毒素攻击后 2小时和 6小时变化显著 ;而CRP在各时间点均变化显著 ,峰值出现在 6小时时间点 ,上升幅度达伤前的 4倍左右。相关分析显示 ,CRP和L PS间呈显著正相关 (r=0 .96 2 1,P<0 .0 1)。结论 :脓毒症大鼠急性期反应显著增强 ,L PS可能具有直接或间接调节
Objective:To analyze the effect of sepsis on the acute phase response and the relationship between synthesis modulation of acute phase reactants and endotoxin.Methods:Sepsis model was replicated by endotoxin administration into the peritoneal cavity in of 40 rats.Animals were randomly divided into normal control,2,6,12,and 24 hours after endotoxin administration groups.Blood samples were collected to determine the concentration of lipopolysaccharide (LPS) with chromogenic limulus amebocyte lysate (LAL) with perchloric acid pretreatment.Creactive protein (CRP) was assayed with the method of turbidimetry.Results:The plasma concentration of LPS increased markedly at 2 and 6 hours groups,while that of CRP increased significantly in all groups after endotoxin challenge,and with 4fold increase in CRP at 6 hours.There was a significant positive correlation between CRP and LPS ( r =0 962 1, P <0 01).Conclusions:Sepsis enhances of acute phase response,and LPS might regulate the synthesis of CRP directly or indirectly.
