摘要
目的 :研究大鼠一过性全脑缺血后 Nestin蛋白在海马中早期表达的变化规律。方法 :雄性 SD大鼠 ,随机分为正常对照组、手术对照组和损伤组。损伤组动物采用四管夹闭脑缺血模型 ,按照脑缺血术后时间不同再分为 1、3、7、14和 2 1日组。应用免疫组织化学法观察 Nestin蛋白及 GFAP蛋白阳性胶质细胞在海马内的变化规律、计数及形态学分析。结果 :细胞计数显示 ,脑缺血后第 1日 Nestin蛋白阳性胶质细胞在海马中出现 ,缺血后第 7日达到高峰之后降低并在缺血第 14日时恢复至对照组水平 ;GFAP蛋白阳性细胞在缺血后第 3日略有增加 ,第 7日达到高峰之后下调。结论
Objective:To investigate the early Nestin expression in hippocampus in rats following the transient cerebral ischemia.Methods:Male SpragueDawley rats were divided into normal control,sham operation control and injury group.Animals in injury group were subjected to four vessels occlusion operation,and then were subdivided into 1,3,7,14 and 21 days groups according to the time elapsed after injury.The expression of Nestin and glial fibrillary acidic protein (GFAP) were determined by immunohistochemistry.Results:The numbers of Nestin positive cells increased bilaterally in hippocampus on day one postinjury,peaked on day seven and then returned to the normal level on day 14 postinjury.The numbers of GFAP positive cells slightly increased on day three,peaked on day seven and then decreased.Conclusions:Nestin is an early sensitive molecular marker for hippocampus injury following transient cerebral ischemia in rats.
