摘要
目的 :将成年大鼠肾上腺皮质细胞分离 ,培养并微囊化后植入肾上腺功能低下大鼠模型体内 ,以探索新的肾上腺移植途径。方法 :成年SD大鼠分成三组 :A组为对照组 ,B组模型植入体外培养并微囊化的肾上腺皮质细胞 ,C组模型植入未经微囊化的肾上腺皮质细胞 ,用放射免疫方法 (RIA)检测移植术后 3 ,9,2 1,42 ,6 3d模型鼠血中皮质醇浓度 ,9周后行组织学检查。结果 :(1)B组大鼠血浆皮质醇最后达 (30 1.33± 88.81)nmol/L ;(2 )植入微囊化肾上腺皮质细胞的模型鼠组织切片 ,未发现排斥反应。结论 :接受微囊化肾上腺皮质细胞移植后肾上腺功能低下大鼠的血清皮质醇水平可接近正常值。
Objective: To find a new way for adrenal transplantation. Methods: Cells from SD rat adrenal cortex were separated, cultured, micro-encapsulated and transplanted into recipient rats with suprarenalectomy. The cortisone in serum were detected by radioimmunoassay (RIA) at 3,9,21,42 and 63 day postoperatively. The pathological changes were observed after 9 weeks. Results: The level of cortisone in serum reached 301.66±88.81nmol/L at 63 day after transplantation. Immunological rejection was not discovered at pathological sections from the transplanted part. Conclusion: The cortisone levels in serum in rats with suprarenalectomy could reach almost normal after micro-encapsulated adrenal cortex cells were transplanted.
