摘要
目的 了解正常人外周血T细胞和胸腺细胞中信号结合T细胞受体删除DNA环 (sjTRECs)的含量 ,从而推测正常人中幼稚T细胞的含量和胸腺的输出功能 .方法 利用实时定量PCR和TaqMan方法检测 11例正常人外周血单个核细胞、7例儿童和 3例成人胸腺细胞DNA中sjTRECs的水平 .结果 正常人中sjTRECs含量分别为 :8.83±4.81/ 10 0 0个外周血单个核细胞 ;2 7.31± 3.2 3/ 10 0 0个成人胸腺细胞和 170 .2 9± 5 9.5 2 / 10 0 0个儿童胸腺细胞 .结论 sjTRECs水平与年龄有关 ,正常人外周血中每 10 0 0个单个核细胞中约含 4.
Objective To analyze the content of signal joint T-cell receptor excision DNA circles(sjTRECs) within peripheral blood mononuclear cells(PBMCs) and thymucytes in normal individuals,thereby to suggest the content of naive T cells and the thymic output function in normal individuals. Methods Quantitative detection of sjTRECs in DNA of peripheral blood mononuclear cells from 11 normal individuals and thymucytes from 7 children and 3 adults were preformed by real-time PCR and TaqMan analysis. Results The mean value of sjTRECs in normal individuals is 8.83+4.81/1000 cells in PBMCs,27.31+3.23/1000 cells in adult thymus and 170.29+59.52/1000 cells in children thymus,respectively. Conclusions The results indicate that the sjTRECs levels may correlate with the age and the number of naive T cells in PBMCs was about 4.5 per 1000 cells.
