八珍汤对辐射损伤小鼠免疫及造血功能的影响 被引量:17

Effects of Bazhen Decoction on Immune Function and Hematopoiesis in Mice with Radiation Damage

摘要 利用 60 Coγ射线 4Gy一次性全身照射造成小鼠骨髓损伤和免疫低下 ,观察体内给药八珍汤(BD)小、中、大 3个剂量后以上功能的恢复情况。实验结果表明 :受照后 ,骨髓中形成CFU -GM集落下降至正常水平的 12 3% ,T、B淋巴细胞增殖能力、NK细胞杀伤活性和TNF、IFN产量明显降低 ,脾脏中CD3+ 、CD4 + 、CD8+ T淋巴细胞 (% )、CD4 + /CD8+ 比值 ,外周血中CD3+ 、CD4 + T淋巴细胞 (% )、CD4 + /CD8+ 比值较正常组升高 ;中、大剂量八珍汤使受照小鼠骨髓CFU-GM集落形成提高至正常的 38%~ 4 1% ;八珍汤 3个剂量组均使T、B淋巴细胞增殖能力恢复至正常水平 ,且呈剂量依赖性增加 ;中剂量八珍汤可使照后小鼠NK细胞杀伤活性提高 ,但还未恢复至正常 ,小、大剂量八珍汤对其NK细胞杀伤活性有抑制作用 ;中剂量八珍汤可降低脾脏中CD3+ 、CD4 + 、CD8+ T淋巴细胞 (% ) ,CD8+ T淋巴细胞百分数恢复至正常 ;小剂量八珍汤降低脾脏中CD3+ 、CD4 + T淋巴细胞百分数 ;脾脏中CD4 + /CD8+ 比值只有八珍汤大剂量降低明显 ,但未恢复至正常 ;3个剂量组八珍汤均使IFN的产量恢复并超过正常水平。结果提示 :八珍汤通过保护免疫器官免受损伤 ,提高淋巴细胞功能及其细胞因子分泌功能 ,来增强机体的细胞免疫功能、体液免疫功能和非特异性免疫功? The effects of small, medium and large doses of Bazhen Decoction (BD) on bone marrow damage and immune depression were investigated in mice suffered from whole body JC60,D,Zo γ ray radiation, 4 Gy for only one time. Compared with the control group, it was found in the radiation group that bone marrow CFU GM colonies decreased to 12 3% of the normal level; T and B lymphocytes proliferation, NK cytotoxic activity and TNF andIFNproductionmarkedlydeclined;splenicCD3 +,CD4 +andCD8 +T lymphocytes percentage and the ratio of CD4 + to CD8 +, and peripheral blood CD3 + and CD4 + T lymphocytes percentage and the ratio of CD4 + to CD8 + increased. The results of BD treatment showed that bone marrow CFU GM colonies were increased by medium and large BD doses to 38%~41% of the normal level; T and B lymphocytes proliferation was restored to the normal level by all the three BD doses in a dose effect manner; NK cytotoxic activity was increased by medium BD dose but not restored to the normal level, but it was further inhibited by small and large BD doses; splenic CD3 +, CD4 + and CD8 + T lymphocytes percentage was decreased by medium BD dose and among them, CD8 + T lymphocytes percentage was restored to the normal level; splenic CD3 + and CD4 + T lymphocytes percentage was decreased by small BD dose; splenic ratio of CD4 + to CD8 + was markedly decreased only by large BD dose, but not restored to the normal level; and IFN production was increased to the normal level and above by all the three BD doses. The results imply that BD enhances cellular immune function, humoral immune function and nonspecific immune function by strengthening lymphocyte function and increasing cytokine production through protecting immune organs from damage, and regulates and controls hematopoiesis through exerting effects on lymphocyte function and cytokine production.
