慢性肝病患者外周血细胞间质胶原酶(MMP-1)及其抑制剂(TIMP-1)基因表达的研究

The expression of MMP - 1 and TIMP - 1 in peripheral mononucleocytes of patients with chronic hepatitis

采用 ＲＴ－ＰＣＲ法及Ｎｏｒｔｈｅｍ杂交，检测 ３８例慢性肝炎患者及 ２０例正常成人外周血单个核细胞中ＭＭＰ－１及ＴＩＭＰ－１ｍＲＮＡ的表达，结果发现正常成人外周血单个核细胞中存在ＭＭＰ－１及ＴＩＭＰ－１的表达，慢肝患者外周血单个该细胞中ＭＭＰ－１及ＴＩＭＰ－１的表达升高，ＭＭＰ－１／ＴＩＭＰ－１比值显著升高，并且与肝纤维化严重程度呈正相关，与肝纤维化血清学指标ＨＡ、ＬＮ、Ｃ－ＩＶ、ＰＣＩＩＩ变化一致。提示外周血单个核细胞中ＭＭＰ－１及 ＴＩＭＰ－１ｍＲＮＡ的表达水平与肝纤维化严重程度关系密切，ＭＭＰ－１／ＴＩＭＰ－１比值可为临床诊断提供一个新的血清学指标。

To study the expression of the metalloproteinase MMP - 1 and its inhibitors TIMP - 1 in peripheral mononucleocytes, 38 patients with chronic hepatitis and 20 normal adults were examined by RT - PCR and Northern blot methods. The results showed that MMP - 1 and TIMP - 1 mRNA were expressed in peripheral monoucleocytes of normal adults, and were highly expressed in peripheral mononucleocytes of patients with chronic hepatitis. The results also indicated that the increase of MMP 1 /TIMP - 1 is positive correlated with the fibrotic staging and necroinflammatory grading in chronic hepatitis and accords with the changes of hepatic firbrosis markers including HA, LN, C- IV and PC III. These date suggest that the level of MMP - 1 and TIMP - 1 are closely associated with hepatic fibrosis development and MMP -1/TIMP- 1 maybe useful in clinic as a novel serum marker.
