正交函数分光光度法测定混合氨基酸的含量

摘要 本文将正交函数分光光度法和茚三酮比色法联用测定混合氨基酸溶液中苯丙氨酸(Phenylalanine,Phe)和天门冬氨酸(Aspartate,Asp)的含量。选用一次正交多项式为特征多项武,波长区间为263~269nm,△λ=2nm的4点法测定Phe的含量,再用比色法测定混合氨基酸的总量,减去Phe的量,可得Asp的量。其平均回收率分别为99.9%,99.3%,变异系数分别为0.5%,0.7%. A mixture of aspartate and phenylalanine was determined quantitatively by means of orthogonal functions spectrophotometry and colorimetry. The first order orthogonal polynomial was selected, the wave length being between 263nm and 269nm, △λ=2nm (n=4). The results showed that the mean recoveries were 99.9% and 99.3% respectively, and the coefficients of variation were 0.5% and 0.7% respectively. This method was simple, rapid and accurate.
