摘要 利用已建立的猪卵母细胞体外无血清培养模型 ,研究了猪卵丘 卵母细胞复合体(CEO)在模拟生理环境的培养条件下Forskolin和促性腺激素对猪卵母细胞成熟的作用。在观察卵母细胞减数分裂恢复 (GVBD )后的实验结果表明 :(1)cAMP产生的激动剂Forskolin(3μmol·L- 1)处理CEO 2 0h可以显著协同HX抑制卵母细胞成熟 ;处理 4 0h后 ,Forskolin(3μmol·L- 1)的抑制作用被卵丘细胞分泌的某种物质克服 ;(2 )Forskolin(3μmol·L- 1)短期 (30 ,6 0 ,12 0 ,2 4 0min)处理后可以诱导卵丘细胞分泌某种促进卵母细胞成熟的物质。 (3)FSH (5 0~ 2 0 0U/L)能够诱导卵丘细胞分泌某种物质促进卵母细胞成熟 ;(4) 5 0U/L的FSH预处理CEO结果与Forskolin结果相同 。 The effect of Forskolin and gonadotropin(FSH) on pig oocyte meiotic resumption were investigated based on the established model of serum free pig oocyte culture in vitro, which could imitate the physiological follicular environment. The GVBD results of CEO showed that (1) Priming pig CEO for 20 hours with Forskolin(3μmol·L -1 ), an activator induced by cAMP, pig oocytes meiotic resumption was inhibited significantly cooperated with HX. After 40 hours' priming, this inhibitory action of Forskolin was overcome by a substance secreted by cumulus cell; (2) When priming pig CEO for short time(30,60,120,240min) with Forskolin(3μmol·L -1 ), cumulus cell could be induced to secrete a substance to stimulate pig oocyte maturation; (3) It is FSH(50~200U/L) that induces cumulus cell to secrete a substance which stimulates pig CEO to resum meiosis; (4) The same result between prepriming pig CEO with 50U/L FSH and with Forskolin suggested that FSH promoting pig oocytes to resume meiosis be mediated by the cAMP path.
