Enhancement of Taxane Production in Suspension Cultures of Taxus chinensis with Elicitation

摘要 通过甲基茉莉酮酸酯 ( MJA)或二氢甲基茉莉酮酸酯 ( HMJA)的诱导 ,有效增强了中国红豆杉 ( Taxus chinensis)悬浮细胞培养中新型生理活性物质云南紫杉烷 ( taxuyunnanine C,Tc)的积累。在培养第 7天添加 5 0 0 μmol/L MJA后 ,产物的胞内最大含量为每克干细胞含 2 9.5 mg Tc,但细胞生长受到明显抑制 ;在添加 1 0 0 μmol/L MJA时 ,Tc的总产量达最大 ,为 30 0 mg/L。 Accumulation of a new physiologically active substance, taxuyunnanine C (Tc), in suspension cultures of Taxus chinensis was enhanced with elicitation of methyl jasmonate (MJA) and dihydro methyl jasmonate (HMJA) on 7th day. The highest Tc content was about 29.5mg/g DW by addition of 500μmol/L MJA, but the cell growth was inhibited in this case. The highest Tc production reached 300mg/L at 100μmol/L of MJA. The activity of a key enzyme, taxadiene synthase, was also greatly increased after elicitation.
