筛选高产变株的最常用的行之有效的手段是上发酵摇瓶,而普通的发酵摇瓶往往只能测得其初始和最终的参数,要想知道发酵过程中的参数变化情况是比较困难的,且取样测定会明显改变发酵液体积。况且取样来测定溶氧是不可靠的。为此。
Shake flasks have been frequently employed for the screening ofhigh productivity strains. However, DO, which is one of the mostimportant parameters, is quite difficult to be monitored in shake flask.culture. A new model of DO probe specially designed for shake flaskculture is of galvanic type. The interior electrode body, which is com-posed of silver (as cathode) and lead (as anode), is inserted in a glassceramic housing. The latter is served as an electrolyte reservior. Theglass ceramic housing can be inserted in a standard ground glass sidetube of 750 ml shake flask. The electrode has been applied to spira-mycin, penicillin fermentation and arthrobacter culture. The informationmonitored by the DO probe proved to be very useful in evaluating theculture process.
