摘要
随着微生物胞内材料在工业和医药方面的应用日益增加,细胞破碎装置的操作也越显得重要。通常采用的菌体处理方法都不同程度地存在局限性,如超声波法和冻融法存在着能源成本问题;机械粉碎法完全破坏了细胞壁,菌体内部物质均被排出,因而给分离带来困难;自溶法。
Aftel high intensity laser irradiation, some microbial intracellularsubstances (e.g.nucleotides) were obtained. This laser technology wassuperior to other cell disintegrated methods, such as sonic disintegrator,freezing and thawing, French pressing, lysozyme and treatment withperchloric acid. Effect of irradiation time, cell concentration and laserpower density on mortality of cells and yield of nucleotides was studied.With the increasing of irradiation time, the mortality of cells and theconcentration of nucleotides exuded out of cells increased.Under certaincell concentration and laser power density (1.25--2.50J/cm^2), the yieldof nucleotides was high.
