摘要
目的 探讨 AST线粒体同工酶 (m- AST)测定的方法学评价。方法 采用选择性抑制法使 s- AST形成免疫复合物后失去活性 ,然后测出余下的 m- AST。结果 该法线性可达到 2 0 0 U/L,重复性试验批内 CV<6 .5 %,批间 CV<9.0 %,回收率 98.2 %~ 1 0 4 .4%,平均为 1 0 1 .4%;血红蛋白≤ 4g/L,胆红素≤3 4 0μmol/L ,甘油三酯≤ 5 .5 mmol/L和抗坏血酸≤ 6 g/L时对 m- AST的活性测定不产生干扰作用 ;初步临床观察 ,其诊断心肌梗死的敏感性优于 AST。结论 m- AST测定的方法简便易行、结果准确可靠 。
Objective To probe into methodology of detection of Mitochondrial aspartate aminotransferase(m AST).Methods The s AST with anti s AST forms antigen antibody complex and loss of activity by select inhibition way,then m AST activity was determined.Results The linearlity of this method was up to 200 U/L The coefficient of variation for within and between assays was less than 6.5% and less than 9.0%.The recoveries range from 98.2% to 104.4% average 101.4%.The m AST doesn't be interference when hemoglobin≤4 g/L,bilirubin≤340 μmol/L,triglyceride≤5.5 mmol/L and vitamine C≤6 g/L Their preliminary applications were founded that the sensitivity of m AST is superior to that of AST for diagnosis of myocardiac infarction.Conclusion The select inhibition for m AST is simple,rapid and reliable,it is one of importance items for clinical diagnosis of myocardiac infarction.
