摘要
目的 :观察大鼠心肌细胞内钙波的影象及钙波在心肌细胞内及细胞间的传播。方法 :采用激光共聚焦显微镜及细胞内钙荧光染色的观测方法。结果 :1在激光共聚焦显微镜下观察了由钙荧光探针 Fluo- 3染色的心房肌及心室肌细胞的影象。 2观察了心肌细胞内钙在某一区域瞬时性增高的现象即钙波在心肌细胞内传播的动态影象 ,钙波的发生与细胞外钙浓度大小有关。3观察了钙波在心肌细胞与细胞间连接处的传播动态影象。结论 :心脏在静息时 (尤其高钙负荷时 )出现钙波是钙信号的一种调节机制。钙波具有在细胞内区域性、瞬时性增高 ,并较快在细胞内传播 ,亦可在细胞间传播。
AIM: To investigate the fundamental nature of calcium waves and its propagation inside cells or in cell-cell junctions. METHODS: We were studied the calcium waves in rat cardiac myocytes loaded with the fluorescent calcium indicator, fluo-3 using confocal laser microscope. RESULTS: ①In the present study, we obtained the images of rat atrial and ventricular myocytes loaded with fluo-3 using confocal laser microscope. ②In the ventricular myocytes, there were local increases of intracellular calcium, i.e. calcium waves, which could be propagated within the cells. The occurrence of calcium waves related to extracellular calcium concentration. ③We also visualized the images of propagation of calcium waves from cell to cell. CONCLUSION: In the heart, waves of calcium release (especially in highly loaded with calcium) are a normal mechanism of cellular signaling. Calcium wave is a regional increase of calcium transient inside the cells, which can propagated within the cells, or somehow between the cells.
