摘要
目的 探讨压力超负荷下心肌细胞凋亡对兔左室重构中的作用。方法 采取末端脱氧核苷酸转换酶介导的dUTP 生物素平移末端标记技术 ,观察持续压力超负荷引起兔心力衰竭的左室心肌细胞凋亡的动态变化。结果 对照组罕见凋亡细胞 ,腹主动脉狭窄术后第 1天凋亡细胞明显高于对照组 (P <0 0 0 1) ,主要分布在心内膜下和乳头肌 ;术后第 3天与术后第 7天之间存在一个峰值 ,凋亡细胞主要分布在心内膜下、乳头肌和心肌中层 ;术后第 14天凋亡数下降 ,但仍与对照组相比存在统计学意义 (P <0 0 5 )。心衰出现后凋亡细胞数再次明显增高 ,与其它各组比较均存在显著性差异 (P <0 0 0 1)。结论 慢性压力超负荷持续存在时 。
Aim To study the feature of myocyte apoptosis on left ventricular remodeling in chronic pressure overload in the rabbit. Methods The models of chronic pressure overload induced heart failure were made by partially ligation of abdominal aorta in 30 rabbits,six normal rabbits served as control. Terminal deoxynucleotidyl transferase mediated dUTP nick end labeling method was used to investigated the dynamic changes of apoptosis in left ventricle myocytes during chronic pressure overload induced heart failure. Result Only a few apoptotic cells can be found in the normal myocardium group. The apoptotic cells number in left ventricle was increased at the first day after operation. Although the apoptotic cells could find in all layers of left ventricle, they were mostly distributed in the subendocardial layers and papillary muscle. The apoptotsis reached its peak at day 3,declined at day 7, and still remained at higher than normal level at day 14. The number of apoptotic cells were the highest in heart failure group ( P <0 001 vs control and no heart failare experiment group). Conclusion Apoptosis may involve in the pathogenesis of heart failure induced by chronic pressure overload.
