摘要
目的探讨高糖血症、高胰岛素血症对 ET- 1缩血管作用的影响及机制。方法复制大鼠高糖血症模型 ,观察高糖组和对照组大鼠的主动脉血管环在 40 m IU/ L 胰岛素中对 10 - 9mol/ L ET- 1的反应 ;测定血浆 NO- 2 含量和血管壁一氧化氮合酶 (NOS)的活性。结果高糖大鼠对 ET- 1的缩血管反应明显强于对照组 (P<0 .0 5 ) ;高糖组NO- 2 含量和 t NOS活性明显低于对照组 ,其中以 c NOS降低为主 (P<0 .0 1)。结论长期高糖血症通过损伤血管内皮功能使 NO合成减少 ,血管对活性物质 ET- 1的反应性增强 ,血管张力增加 。
Objective To investigate the effects of hyperglycemia an d h yperinsulinism on vasoconstriction caused by endothelin-1(ET-1).Methods Hyperglycemia model rats were prepared.We observed the responses of aorta rings obtained from hyperglycemic and normal rats to 10 -9 mol/L ET-1 in 4 0 mIU/L insulin solution and determined contents of NO - 2 and NOS.Resu lts Rings constricted more obviously in normal rats than those in hyperglycemic rat s ( P <0 05) and the contents of NO and NOS were lower in hyperglycemic rats .Conclusion Hyperglycemia could damage vascular endothelium and enha nce vascular tension.This may be involved in pathogenesis of vascular complicati on,such as atherosclerosis.\;
